11.  Composition identification and functional verification of bacterial community in disease-suppressive soils by machine learning.LinkIT

However, the general composition and functional profiles of bacterial community in soils which suppress multiple diseases for various plants remain largely unknown. Here, we combined metadata analysis with machine learning to identify the general patterns of bacterial-community composition in disease-suppressive soils. Disease-suppressive soils significantly enriched Firmicutes and Actinobacteria but showed a decrease in Proteobacteria and Bacteroidetes. Our machine-learning models accurately identified the disease-conducive and -suppressive soils with 54 biomarker genera, 28 of which were potentially beneficial. We further carried out a successive passaging experiment with the susceptible rps2 mutant of Arabidopsis thaliana invaded by Pseudomonas syringae pv. tomato DC3000 (avrRpt2) for functional verification of potential beneficial bacteria. The disease-suppressive ability of Kribbella, Nocardioides and Bacillus was confirmed, and they positively activated the pathogen-associated molecular patterns-triggered immunity pathway. Results also showed that chemical control by pesticides in agricultural production decreased the disease-suppressive ability of soil. The samples were analyzed regarding the impact of environmental factors on the composition of apocarotenoids and phenolic constituents. Quantification of saffron metabolites was carried out using high-performance liquid chromatography. It was found that the high content of chlorogenic acid (0.2?mg/g, Ukraine) and ferulic acid (0.28?mg/g, India) was controlled by the duration of solar radiation during plant development. The accumulation of caffeic acid (the higher content 4.88?mg/g, Ukraine) in stigmas depended on the average air temperature. In contrast, the total crocins content according to the correlation analysis depended on the duration of solar radiation, the solar UV index, and the soil type. Rutin was found in all samples (0.83-8.74?mg/g). The highest amount of crocins (average 382.45?mg/g) accumulated in saffron from Italy and Ukraine. Crocins, picrocrocin, safranal, and rutin can further serve as saffron quality markers. All validation parameters were satisfactory and high-performance liquid chromatography methods could be successfully applied for the composition assessment of saffron metabolites. Saffron extracts showed the highest antibacterial activity against Bacillus subtilis, Staphylococcus aureus, and Escherichia coli (MICs 62.5-125?µg/ml).© 2022 Deutsche Pharmazeutische study was aimed to designed laboratory scale constructed wetland (CW) for the treatment of domestic wastewater under temperature range (18-38°C) and hydraulic retention times (24 and 48?h). Besides, the soil of vegetative unit of CW was assessed using conventional culturing techniques, and 13 different bacterial species (Escherichia coli, Micrococcus, Pseudomonas, Proteus, Klebsiella, Streptococcus, Alcaligenes, Salmonella, Bacillus, Enterobacter, Staphylococcus, Shigella and Corynebacterium spp.) were determined. The pathogenic microbial load was high in influent samples, but after treatment, about 73.1-99.7% and 43.5-86.7% reduction in CFU/ml and MPN/100?ml index, respectively, were observed. Moreover, the organic loads in terms of COD, TDS, TSS, and turbidity were high in all influent samples, but after treatment, average percentage removal in different physico-chemical parameters was observed during overall treatment operations, that is, COD (59.7-65.6%), TDS (59.6-76.8%), TSS (64.9-76.7%), and turbidity (72.7-91.6%), while pH of the effluent samples was observed in the prescribed limits. It was concluded that laboratory scale CW using natural flora Paspalidium flavidum and bacterial species was efficient in the reduction of different pollution indicators and hence a best option to be modified on pilot-scale for wastewater treatment in the rural regions of Peshawar. PRACTITIONER POINTS: Role of retention times on performance of CW were studied. The commonly existing vegetation was utilized to treat domestic waste water.
